There is a "bus station" (terminal) in Siem Reap, where interstate buses depart and arrive. Normally the bus company will arrange a pick up for you from your accommodation and will drop you at their downtown office (about 30 minutes prior to the scheduled departure time). Once everybody has been gathered at the company's branch in downtown Siem Reap, then they will be transferred to this bus station, where buses will actually depart.
Arriving in Siem Reap, Remorks (Tuk-tuk) are normally available around the bus station; alternatively your hotel should be able to arrange a pick up for you. If you are travelling by a "Ford Minivan", you might be able to alight in downtown Siem Reap. See below for information on minivans.
Bus is a good option to reach Siem Reap from Phnom Penh. Starting price is around US$ 4-5 for an air-conditioned bus without toilet and around $ 6 for the one with a lavatory. Tickets can be purchased directly at bus companies or through travel agents in the city (they may charge an additional fee of approximately $ 1). These fares normally include a light snack and a small glass/bottle of mineral water.
Usually it takes around 6 hours to reach Siem Reap from Phnom Penh and vice versa, since road conditions are today generally good.
The so-called "Ford minivan" is available for travelers who wish to reach their destination slightly faster than buses, by about 30 minutes to 60 minutes. The minivan accommodates up to 15 people and costs around $ 9. It may stop at a restaurant in a town called Kampong Thom for a 20-minute break, though travelers might be told that it would be a non-stop journey when they buy their tickets.
If you choose to take this minivan you should avoid the most aft seats, unless you like enjoying a bumpy ride, and if you need more space for your legs, don't take the window seat at this most aft row because the minivan's wheels already take up a bit of the space.

By Shared Taxi
There are "shared taxi" services (using Toyota Camrys, don't expect to see brand new ones) from one town to another town in Cambodia. This type of taxi is usually shared by 6-7 passengers, excluding the driver. Though it is more expensive than interstate buses, you can reach your destination faster, however it is more comfortable travelling by bus, since you will have more space for yourself and more space for your belongings.
A shared taxi trip from Siem Reap to Phnom Penh takes about 4.5 hours at a cost of around 50,000 Riel, to Sihanoukville is around 30,000 Riel (about 4 hours journey). They leave early from around 5am and last departure is usually around 3pm. Guesthouses in general know how to get one for you, but probably you need to compare their fares and details to avoid being overcharged or misinformed.
